Lipoteichoic acid (LTA) is a major amphiphilic polymer found in the cell wall of Gram-positive bacteria. It is anchored to the cytoplasmic membrane via a glycolipid and extends through the thick peptidoglycan layer characteristic of these organisms. LTA plays crucial roles in bacterial physiology, immune modulation, and pathogenesis. It binds to host receptors like CD14 and Toll-like receptor 2 (TLR2), activating NF-kB signaling pathways and mitogen-activated protein kinases (MAPK), leading to changes in gene expression related to inflammation and apoptosis regulation. Enzymes involved in its synthesis are potential targets for novel antimicrobials or combination therapies against resistant pathogens.
