Overview

Liprin-beta-1, encoded by the *PPFIBP1* gene, is a scaffold protein belonging to the liprin family. Liprin proteins, including liprin-beta-1, interact with members of the LAR family of transmembrane receptor protein tyrosine phosphatases, which are important for axon guidance, mammary gland development, and cell adhesion[3][5]. Liprin-beta-1 forms homo- and hetero-dimers, contributing to the structural organization at focal adhesions and within neuronal synapses[1][6]. It is essential for neuronal outgrowth and synapse formation, as shown in model organisms including Drosophila and C. elegans, with loss-of-function mutations causing profound neurodevelopmental disorders characterized by epilepsy, microcephaly, abnormal vision, and structural brain abnormalities[4][6]. Fusion events involving *PPFIBP1* have been identified in certain cancer subtypes, suggesting a possible oncogenic potential[1]. However, PPFIBP1/liprin-beta-1 itself is not a receptor, enzyme, transporter, or direct drug target, but rather acts as a scaffold facilitating cellular localization and interaction of other proteins, especially within the nervous system and during cell adhesion processes[1][3][6].
