Overview

Liver cancer cell metastasis is a complex, multi-step biological process characterized by the spread of malignant cells from the primary liver tumor to distant sites such as the lungs, bones, or lymph nodes (National Cancer Institute, 2023). It is not a single molecular target (like a protein or receptor) but rather a phenotypic progression of disease involving the dysregulation of multiple signaling pathways, including MET, VEGFR, and TGF-beta (PubMed: 32805445). During this process, cancer cells undergo epithelial-mesenchymal transition (EMT), allowing them to degrade the extracellular matrix, enter the bloodstream (intravasation), and eventually colonize secondary organs. Therapeutic interventions aimed at metastasis typically involve multi-kinase inhibitors or monoclonal antibodies that target the underlying growth factor receptors and angiogenic signals that facilitate cell spread (NIH, 2022). Because metastasis involves systemic interactions between tumor cells and the microenvironment, it represents a significant challenge for clinical management and is the leading cause of mortality in patients with hepatocellular carcinoma.
