Overview

Liver cell receptor is a broad classification encompassing numerous distinct proteins expressed on the surface or within the cytoplasm of hepatic cells, such as hepatocytes, Kupffer cells, and stellate cells (StatPearls, 2023). These receptors include the asialoglycoprotein receptor (ASGPR), which is highly expressed on hepatocytes and mediates the endocytosis of glycoproteins (UniProt P07306), and the farnesoid X receptor (FXR), a nuclear receptor critical for bile acid and lipid homeostasis (PubMed: 10331394). Collectively, these receptors regulate vital physiological processes including glucose metabolism, detoxification, and immune responses within the hepatic microenvironment (NIH, 2022). Because the term refers to a heterogeneous group of molecules rather than a single protein, it is considered a generic category rather than a specific therapeutic target. However, specific members of this group are targeted by drugs like obeticholic acid for primary biliary cholangitis or utilized for targeted delivery of RNA therapeutics using GalNAc ligands (Nature Reviews Drug Discovery, 2020). Dysregulation of these various receptors is implicated in the progression of chronic liver diseases, including non-alcoholic steatohepatitis (NASH) and hepatocellular carcinoma (PubMed: 31535004).
