LLGL scribble cell polarity complex component 1 (LLGL1, commonly known as Lethal giant larvae homolog 1 or Lgl1) is a highly conserved protein that functions as part of the Scribble polarity complex, together with Scribble (Scrib) and Discs large (Dlg)[1][2][3][5]. This complex is essential for the establishment and maintenance of cell polarity, cell adhesion, and directed cell migration in a broad range of tissues and organisms, from fruit flies to mammals[1][3][5]. LLGL1 acts as a scaffold or adaptor protein, interacting with other polarity proteins such as Scrib and Dlg to regulate the assembly and stability of multi-protein complexes crucial for apicobasal polarity and tissue architecture[1][3]. Loss or decreased expression of LLGL1 has been associated with loss of cell polarity, increased cell migration, tissue disorganization, and neoplastic transformation (acting as a tumor suppressor)[1][3][5]. In carcinomas, the Scrib-Lgl-Dlg module is frequently disrupted, contributing to the development and progression of cancer[1][3][5]. LLGL1 also plays a role in the regulation of cell junctions, including both adherens and tight junctions, which impact tissue barrier functions[2][3]. **LLGL1 is not a classical therapeutic target** such as a receptor, enzyme, or transporter. No drugs are currently known to directly interact with it.
