01

Overview

LMNTD2 antisense RNA 1 (LMNTD2-AS1) is a human long non-coding RNA (lncRNA) gene located on chromosome 11p15.5. It acts as an antisense transcript regulating the LMNTD2 gene and other targets by transcriptional interference, recruitment of transcription factors, and influencing alternative splicing. LMNTD2-AS1 has emerged as an oncogenic lncRNA markedly upregulated in several cancers, most notably prostate cancer, correlating with tumor progression, poor prognosis, and increased immune cell infiltration in the tumor microenvironment. Its molecular functions include the regulation of gene expression via epigenetic and transcriptional control mechanisms. LMNTD2-AS1 can interact with RNA-binding proteins such as FUS, potentially modulating pathways like NRF2 signaling. LMNTD2-AS1 is being studied as a biomarker for cancer diagnosis and prognosis but has not yet been directly targeted by any approved therapeutic agents.

Mechanism of action

Not applicable; as no drug is described to target LMNTD2-AS1 directly, mechanisms of action are not established. Potential mechanisms under investigation could involve modulation of the NRF2 pathway or immune cell infiltration.

03

Biological functions

Regulation of gene expression (by antisense interaction and promoter targeting)Regulation of alternative splicingRecruitment of transcription factorsRegulation of immune cell infiltration in the tumor microenvironmentPromotion of cell proliferation, migration, and invasion in cancerRegulation of the NRF2 signaling pathway (via FUS protein binding)
04

Disease associations

Cancer (with demonstrated roles in prostate cancer and differential expression in e.g. cholangiocarcinoma, kidney cancer, bladder cancer, cervical cancer, prostate adenocarcinoma, breast cancer, lung adenocarcinoma)Immune response and tumor progression (by influencing immune cell infiltration)
05

Safety considerations

Not establishedAs with other lncRNA therapies, general concerns might include off-target gene regulation and tissue specificity
06

Biomarkers

LMNTD2-AS1 itself is a candidate biomarker for cancer, especially prostate cancer, as its elevated expression is associated with poor prognosis and can differentiate tumor from normal tissue (AUC=0.767 for prostate cancer)
