LncRNA Neighboring Enhancer of FOXA2 (LNCNEF)

Target
LNCNEF
Molecular classification
Long non-coding RNA (lncRNA), member of the regulatory non-coding RNA family, Not a protein, receptor, enzyme, transporter, or transcription factor, but a regulatory RNA
01

Overview

LncRNA Neighboring Enhancer of FOXA2 (LNCNEF) is a long non-coding RNA molecule involved in the modulation of gene expression by enhancer-like mechanisms, particularly influencing FOXA2 and pathways such as TGF-β signaling. It acts as a tumor suppressor in certain cancers, notably hepatocellular carcinoma and small cell lung cancer, where its downregulation is associated with metastasis and poor prognosis. In ankylosing spondylitis, LNCNEF is upregulated and correlated with disease activity and recurrence, making it a novel candidate biomarker for disease monitoring. LNCNEF represents a potential target for therapeutic modulation, though direct targeting in drugs has not been achieved, and further research is needed to clarify its molecular functions and clinical utility.

Mechanism of action

For nonsteroidal anti-inflammatory drugs: indirect downregulation of LNCNEF by reducing inflammation. For potential future drugs: hypothetical interference with lncRNA expression or function, not yet established.

03

Biological functions

Regulation of gene expression, especially local enhancer effects on FOXA2 and potentially other genesTumor suppression in hepatocellular carcinoma and small cell lung cancer through modulation of cell migration and invasionModulation of inflammatory response in synovial fibroblast-like cells and regulation of inflammatory cytokine expressionInvolvement in epithelial-mesenchymal transition (EMT), especially by influencing the TGF-β signaling pathway
04

Disease associations

Cancer (tumor suppressor in hepatocellular carcinoma, diagnostic/prognostic marker in small cell lung cancer)Ankylosing spondylitis (prognostic biomarker for recurrence and treatment outcomes)Potential association with osteoporosisThyroid cancer, nonmedullary (genetic association)
05

Safety considerations

No safety concerns specific to LNCNEF are documentedgeneral challenges include delivery of RNA-targeted therapeutics and off-target effects typical of nucleic acid therapiesClinical impact as a therapeutic target is still under investigation
06

Interacting drugs

Nonsteroidal anti-inflammatory drugs (NSAIDs)
07

Biomarkers

LNCNEF itself is proposed as a biomarker for recurrence and poor outcomes in ankylosing spondylitisPlasma and tissue LNCNEF levels as diagnostic/prognostic indicators in small cell lung cancer
