Overview

LncRNA periodontal mesenchymal stem cell osteogenesis related (LNCPOIR) is a long non-coding RNA (lncRNA) specifically implicated in the regulation of osteogenic differentiation in periodontal mesenchymal stem cells (PDLSCs). Identified as lncRNA00638 (also referred in databases as ENSG00000232395 and NCBI Gene ID: 105377959), LNCPOIR is significantly upregulated during the process of osteogenic induction in PDLSCs, especially under static mechanical strain conditions present in periodontal inflammation. Mechanistically, LNCPOIR acts as a competing endogenous RNA (ceRNA) for miR-424-5p, thereby regulating the expression of fibroblast growth factor receptor 1 (FGFR1), which is involved in bone formation and remodeling. Experimental studies have shown that overexpression of LNCPOIR promotes osteogenic differentiation, evidenced by increased expression of osteogenesis markers (like Runx2, ALP, Col1), increased alkaline phosphatase activity, and bone matrix mineralization. Downregulation impairs these processes. LNCPOIR is regarded as a promising therapeutic target for improving bone regeneration and optimizing orthodontic therapy in patients with periodontitis, though it is not a classic druggable protein target. No direct drug modulators of LNCPOIR are currently described.
