Overview

Local immune tolerance induction refers to the mechanisms by which the immune system actively suppresses or limits immune responses in specific tissues or microenvironments, preventing unwanted inflammation and autoimmunity while maintaining tissue homeostasis. This process is crucial for protecting normal tissues from immune-mediated damage, especially at barrier sites such as the gut, skin, and mucosal surfaces. Key cellular players include regulatory T cells (Tregs), tolerogenic dendritic cells (DCs), and macrophages. Molecular mechanisms involve Foxp3, IL-10, IDO, retinoic acid, and the Sialic Acid–Siglec Axis. Defects in these mechanisms contribute to various diseases, making them therapeutic targets for enhancement (in autoimmunity/allergy) or breakdown (in cancer).
