Overview

Local tissue inflammation mediators are a diverse group of molecules produced at sites of tissue injury or infection that orchestrate the inflammatory response. These mediators are released by various cells, including leukocytes, macrophages, endothelial cells, and platelets, as well as from plasma-derived systems. They act through specific receptors to induce vasodilation, increased vascular permeability, chemotaxis, pain, and leukocyte activation, ultimately contributing to the signs and symptoms of inflammation. Dysregulation of these mediators underlies many chronic diseases.

Mechanism of action

Varies depending on the specific mediator and drug. Examples include: Inhibition of prostaglandin synthesis (NSAIDs); suppression of cytokine production (corticosteroids); blocking histamine receptors (antihistamines); neutralizing TNF-alpha (anti-TNF agents); blocking leukotriene receptors or synthesis (leukotriene inhibitors).
