Lolium perenne pollen allergen Lol p 5 is a major protein allergen derived from perennial ryegrass, a primary source of seasonal aeroallergens globally (UniProt Q40240). It belongs to the Group 5 grass pollen allergens, which are characterized by their high solubility and potency in inducing IgE-mediated Type I hypersensitivity (IUIS Allergen Nomenclature). Lol p 5 is localized within the starch granules of pollen and is released upon contact with mucosal surfaces. Once released, it cross-links IgE on mast cells and basophils, triggering the release of inflammatory mediators like histamine (PubMed: 8230054). In clinical practice, Lol p 5 is a critical component of diagnostic tests and allergen-specific immunotherapy (AIT) products. Therapeutic strategies involving Lol p 5 include the use of standardized grass pollen extracts to desensitize patients. The therapeutic goal of targeting Lol p 5 via AIT is to shift the immune system from a Th2-dominated allergic response to a Th1/Treg-mediated tolerant state (PubMed: 25173434). This modulation leads to a reduction in symptoms of allergic rhinitis and asthma. Monitoring of therapy often involves measuring changes in allergen-specific IgE and IgG4 levels. Safety remains a concern during treatment due to the risk of systemic allergic reactions or anaphylaxis.
