Long intergenic non-protein coding RNA 1010 (LINC01010) is a cell-type specific long non-coding RNA (lncRNA) predominantly expressed in myeloid cells, such as macrophages[2][4]. It is upregulated during differentiation and polarization of both M1 and M2 macrophage subsets and shows dynamic regulation in response to bacterial stimuli, especially in the context of periodontal disease[2][4]. Functionally, LINC01010 is essential for the acquisition of key innate immune activities, including expression of differentiation and polarization markers (such as CD206, CD68, MHCII, CD32, CD209), effective phagocytosis of bacteria, antigen processing, and secretion of pro-inflammatory cytokines (IL-1β, IL-6, IL-8, TNF-α). Knockdown of LINC01010 has been shown to impair these innate immune processes, in part by suppressing expression of genes in the NFκB signaling pathway, indicating a regulatory role in macrophage-mediated immunity. LINC01010’s dysregulation is associated with immune dysfunction in periodontal disease, highlighting its significance as both a functional RNA and a potential disease biomarker[2][4].
