Long intergenic non-protein coding RNA 1016 (LINC01016) is a long non-coding RNA (lncRNA) gene located on chromosome 6q21.31 and is expressed as multiple isoforms, particularly abundant in testis and variably expressed in several tissues and cancers[1][5]. LINC01016 has been mechanistically linked to the promotion of cell proliferation, migration, and invasion in several cancers, including gastric, breast, endometrial, testicular, and cervical cancer. Its oncogenic effects are mediated through various mechanisms, such as binding to proteins (e.g., binds EIF4A3, thereby regulating nonsense-mediated RNA decay and increasing MMP9 expression, or DHX9, stabilizing it and enhancing signaling pathways like PI3K/AKT)[1][2][3]. LINC01016 expression correlates with poor prognosis, metastatic potential, and relapse in gastric cancer and other tumors, making it a putative therapeutic and diagnostic target. However, there are currently no known drugs that directly target LINC01016, and its context-specific isoform expression poses additional therapeutic challenges[5].
