Long intergenic non-protein coding RNA 1048 (LINC01048) is a human long non-coding RNA transcribed from an intergenic region on chromosome 13[1][5]. It has been identified as an oncogenic lncRNA with elevated expression in multiple cancers, particularly gastric cancer and cutaneous squamous cell carcinoma. Mechanistically, LINC01048 promotes cell proliferation and suppresses apoptosis by modulating major oncogenic signaling pathways: it enhances the nuclear localization and activity of β-catenin (implicating it in the Wnt/β-catenin pathway) in gastric cancer[1], and upregulates YAP1 via interaction with the transcription factor TAF15 within the Hippo pathway in cutaneous squamous cell carcinoma[2][3]. Knockdown of LINC01048 suppresses tumor cell growth and increases apoptotic rates in multiple in vitro and in vivo models, highlighting its candidacy as a biomarker and potential therapeutic target in cancer. High levels of LINC01048 are consistently correlated with poorer prognosis in patient studies[1][2][3][5]. No direct pharmacological inhibitors exist, but pathway modulators like SKL2001 can counteract the effects of LINC01048 knockdown in preclinical models[1].
