Overview

Long intergenic non-protein coding RNA 1111 (LINC01111) is a long non-coding RNA (lncRNA) located at chromosome 8q21.13 that does not encode a protein. LINC01111 is notably downregulated in pancreatic cancer tissues and the plasma of affected patients. It exerts tumor-suppressive effects primarily by acting as a competing endogenous RNA (ceRNA), sponging miR-3924 and thereby upregulating dual-specificity phosphatase 1 (DUSP1), which inhibits the stress-activated protein kinase/c-Jun N-terminal kinase (SAPK/JNK) pathway. Through these actions, LINC01111 inhibits cell proliferation, cell cycle progression, invasion, and migration of pancreatic cancer cells in vitro, and suppresses tumorigenesis and metastasis in vivo. Expression of LINC01111 is negatively correlated with tumor stage and positively correlated with patient survival, suggesting it is a potential diagnostic and prognostic biomarker in pancreatic cancer[1][2][4][5][6][7]. LINC01111 is not classified as a classical therapeutic target (such as a receptor, enzyme, transporter, or ion channel) but is an important regulatory molecule in cancer biology and a potential biomarker. No drugs or direct molecular inhibitors/activators of LINC01111 are currently described in the literature, and it does not serve as a direct drug target[1][2][3][5].
