Overview

**Long intergenic non-protein coding RNA 1117 (LINC01117)** is a long non-coding RNA belonging to the subclass of long intergenic non-coding RNAs, defined as RNA molecules longer than 200 nucleotides and located between protein-coding genes[1][3]. LINC01117 is highly expressed in lung adenocarcinoma (LUAD) cells and tissues and plays a significant role in promoting migration and invasion of cancer cells[2][4][6]. Mechanistically, LINC01117 regulates the epithelial–mesenchymal transition (EMT) process, largely by modulating the nuclear and cytoplasmic distribution of YAP1, a key effector of the Hippo signaling pathway[2][4][6]. Knockdown of LINC01117 suppresses EMT and reduces cell motility, while overexpression enhances pro-metastatic features. Its expression is associated with poor prognosis in lung adenocarcinoma patients, suggesting its potential utility as a prognostic biomarker and as a potential non-coding RNA therapeutic target. There are currently no known drugs that directly target LINC01117, but its functional roles make it an emerging focus for research into RNA-based cancer therapeutics.

Mechanism of action

Not applicable for classical small molecule or antibody targeting; functionally, LINC01117 can be silenced by RNA interference (siRNA) or modulated by overexpression systems in research contexts

03

Biological functions

Regulation of epithelial–mesenchymal transition (EMT)Regulation of cell migration and invasionModulation of Hippo pathway signaling (influencing YAP1 nuclear/cytoplasmic distribution)Likely involved in gene regulation and chromatin architecture (general for lincRNAs)
04

Disease associations

Cancer (specifically lung adenocarcinoma)
05

Safety considerations

Potential safety challenges regarding the specificity and off-target effects of lincRNA-targeting therapeutics are inherent to all long non-coding RNAs, but no specific safety data are reported for LINC01117
06

Biomarkers

Prognostic biomarker for lung adenocarcinoma (high expression correlated with poor prognosis and advanced clinical features)
