Overview

LINC01127 is a long intergenic non-coding RNA identified primarily in human glioblastoma and other cancers as a regulatory molecule with roles in gene regulation and cellular phenotype modulation. It is found in regions of the human genome that do not overlap coding genes. It regulates gene expression, notably self-renewal and stemness in glioblastoma cells, through chromatin remodeling and enhancing transcription of the MAP4K4 gene in a cis-regulatory manner. It is upregulated by histone lactylation associated with metabolic reprogramming (Warburg effect) and acts through the MAP4K4/JNK/NF-κB pathway. High LINC01127 expression is correlated with poor prognosis in glioblastoma, and targeting this axis impedes tumor growth in preclinical models, suggesting its potential as a therapeutic target and biomarker.

Mechanism of action

LINC01127 acts through cis-regulation of MAP4K4 by guiding POLR2A to promoter regions, activating the JNK pathway and modulating self-renewal of glioblastoma cells. It is upregulated by histone lactylation induced via the Warburg effect, which activates NF-κB signaling. Targeting its axis/mechanism (e.g., with curcumin for NF-κB inhibition) has been studied preclinically.
