Overview

Long intergenic non-protein coding RNA 1134 (LINC01134), also known as TLNC1 (tumorigenic long noncoding RNA on chromosome 1p13), is an oncogenic long non-coding RNA strongly upregulated in hepatocellular carcinoma (HCC)[1][3][5]. It regulates multiple biological processes relevant to tumorigenesis and cancer progression, notably by promoting cell proliferation, migration, invasion, and resistance to therapy. Mechanistically, LINC01134 acts as a molecular scaffold, microRNA (miRNA) sponge, and modulator of chromatin or protein complexes. It enhances tumor cell aggressiveness and therapy resistance by repressing tumor suppressor signals such as the p53 pathway (via TPR-mediated nuclear export), activating oncogenic signaling pathways (NF-κB, MAPK), and conferring resistance to chemotherapeutic agents and radiotherapy[1][3][5]. LINC01134 expression is associated with poor prognosis and is a potential biomarker and therapeutic target in liver cancer, although as of current literature, there are no approved direct inhibitors or drugs targeting this molecule in clinical use[3][5].
