Long intergenic non-protein coding RNA 1144 (LINC01144) is a long noncoding RNA (lncRNA) that is not translated into protein but plays regulatory roles in gene expression. LINC01144 is upregulated in colorectal cancer tissues and cell lines and is associated with tumor progression. It functions mechanistically as a competing endogenous RNA (ceRNA), sequestering microRNA-331 (miR-331), resulting in increased levels of HER2 (a recognized oncogenic receptor tyrosine kinase), as well as heightened Akt and ERK1/2 signaling, thus promoting proliferation and invasion of colorectal cancer cells. Silencing LINC01144 suppresses proliferation, reduces invasion, and increases apoptosis in colorectal cancer cell models. Its apparent disease relevance is so far limited primarily to colorectal cancer, with evidence supporting its role as a biomarker and possible therapeutic target in this malignancy, but there are no known drugs targeting LINC01144 or established clinical safety concerns associated with its modulation.
