Overview

Long intergenic non-protein coding RNA 1305 is a non-coding RNA transcript (3122 bp, three exons), located on the forward strand of chromosome 2. It does not encode a protein but is primarily expressed in the cytosol and exosomes of tumor cells. LINC01305 is notably upregulated in cervical cancer and esophageal squamous cell carcinoma tissues, where it promotes cell proliferation, migration, invasion, and inhibits apoptosis. It exerts its effects by interacting with RNA-binding proteins (such as KHSRP and BNC1) and regulates critical signaling pathways (JNK and Wnt) related to EMT, stemness, and tumor progression. High expression of LINC01305 is associated with poor clinical prognosis and may serve both as a biomarker and as a potential therapeutic target in oncology.

Mechanism of action

When targeted, inhibition or knockdown of LINC01305 leads to reduced tumor growth, decreased proliferation, and increased apoptosis. Its oncogenic effects are mediated by interaction with protein partners (KHSRP, BNC1), impacting transcriptional and signaling pathways (e.g., PI3K/Akt, JNK/EMT).

Safety considerations

Major safety concerns are not reported for targeting LINC01305, but therapeutic targeting of lncRNAs in cancer biology may encounter challenges such as cellular delivery, off-target effects, and specificity

Biomarkers

LINC01305 expression levels serve as biomarkers for poor prognosis in cervical cancer; its upregulation is correlated with reduced survival and aggressive tumor phenotypes
