Long intergenic non-protein coding RNA 1354 (LINC01354) is a cytoplasmic long non-coding RNA of 944 nucleotides located on chromosome 1. It is upregulated in several human cancers, including gastric cancer, colorectal cancer, lung cancer, and osteosarcoma. Functionally, LINC01354 acts as an oncogene, promoting tumor cell proliferation, migration, invasion, and epithelial-mesenchymal transition—largely via two distinct molecular mechanisms: - acting as a competing endogenous RNA (ceRNA) for microRNAs (for example, sponging miR-153-5p to regulate CADM2 in gastric cancer, or miR-340-5p/ATF1 signaling in lung cancer), - stabilizing mRNA transcripts (stabilizing β-catenin mRNA through interaction with hnRNP-D, thereby activating the Wnt/β-catenin signaling pathway in colorectal cancer). Overexpression of LINC01354 correlates with poor prognosis, increased tumor size, metastasis, and advanced clinical stage in several cancer types, making its expression a potential biomarker for diagnosis and prognosis. Current research focuses on its functional role in tumor biology; no marketed therapeutics target LINC01354 directly at this time.
