Long intergenic non-protein coding RNA 1446 (LINC01446) is a human long non-coding RNA located at chromosome 7p12.1 and spans 3,484 nucleotides. LINC01446 is frequently upregulated in several cancers, most notably gastric cancer, where its overexpression correlates with poor patient prognosis and increased tumor aggressiveness. Functional studies demonstrate that LINC01446 promotes tumor cell proliferation, migration, invasion, and suppresses apoptosis, both in vitro and in vivo. Mechanistically, LINC01446 acts partly by binding to the histone lysine-specific demethylase LSD1, recruiting it to the promoter of the RASD1 gene and causing its transcriptional downregulation via epigenetic H3K4me2 demethylation. This LINC01446/LSD1/RASD1 regulatory axis is implicated in oncogenesis and may provide therapeutic opportunities for targeting advanced gastric cancer and potentially other tumor types. No direct drug interactions are currently reported, but its classification as an oncogenic lncRNA makes it a promising molecular target for further investigation.
