Overview

Long intergenic non-protein coding RNA 1488 (LINC01488) is a non-coding RNA located at chromosome 11q13.3 (3425 bp) and functions as a tumor suppressor in liver and breast cancer. It inhibits cell proliferation and tumor metastasis by binding and promoting the ubiquitination of cyclin E and modulating the miR-124-3p/miR-138-5p/vimentin axis. High levels of LINC01488 correlate with improved cancer prognosis and survival, making it a promising biomarker and potential therapeutic target in oncology.

Mechanism of action

Biological regulation is via: Transcriptional repression of cyclin D1 via TLS binding and inhibition of histone acetyltransferase activity; Ubiquitination and degradation of cyclin E; Modulation of miRNA biogenesis for miR-124-3p and miR-138-5p, reducing vimentin expression.

03

Biological functions

Cell proliferation (negatively regulates proliferation in hepatocellular carcinoma)Apoptosis (involved in apoptosis regulation)Cell cycle (modulates cyclin E, cyclin D, CDK2, and CDK4 protein levels)Metastasis (suppresses metastasis via the miR-124-3p/miR-138-5p/vimentin axis)DNA repair (involved in DNA double-strand break repair pathway modulation in breast cancer)
04

Disease associations

Cancer (tumor suppressor in hepatocellular carcinoma, implicated as a biomarker for breast cancer subtypes)Other (possible roles in other tumor types are under study, but cancer is currently the major disease context)
05

Safety considerations

No direct therapeutic agents reported; thus, no documented safety concerns or off-target effects.Overexpression studies suggest that increasing LINC01488 suppresses cell proliferation and metastasis without known adverse effects in preclinical models.
06

Biomarkers

Expression level of LINC01488 is proposed as a prognostic biomarker in hepatocellular carcinoma and as a subclassifier for breast cancer
