Overview

LINC01518 (long intergenic non-protein coding RNA 1518), also called JINR1 (JEV-induced non-coding RNA 1), is a primate-conserved lncRNA highly expressed in various cancers (including head and neck, esophageal, prostate, and bladder cancers) and upregulated upon certain viral infections, particularly neurotropic flaviviruses like Japanese encephalitis virus and West Nile virus[1][2][3][4][5][6]. It acts as a ceRNA by sponging microRNAs such as miR-1-3p, miR-216b-5p, and miR-320a, which leads to increased expression of target genes involved in proliferation, migration, autophagy, and resistance to apoptosis (notably Slug, GRP78, DDX5, and CNKSR2)[3][5][6]. LINC01518 promotes tumor growth and metastasis, enhances viral replication and inflammation-induced cell death, and serves as a biomarker for prognosis in prostate cancer and possibly other diseases[2][3][5][6]. There are currently no approved drugs targeting LINC01518 directly, but knockdown studies suggest therapeutic potential.

Mechanism of action

Cisplatin: induces apoptosis in HNSCC cells, with enhanced sensitivity when LINC01518 is depleted. No direct mechanism for targeted small molecules or biologics, as LINC01518 operates by sponging specific miRNAs.
