Overview

Long intergenic non-protein coding RNA 1546 (LINC01546, also known as VAL or CXorf28) is a cytoplasmic lncRNA directly induced by AKT/STAT3 signaling and essential for tumor dissemination in lung adenocarcinoma. Unlike other lncRNAs that drive epithelial-mesenchymal transition, VAL mediates metastasis independently of EMT by binding Vimentin and preventing its TRIM16-mediated polyubiquitination and degradation. This results in stabilization of Vimentin, increased cell invasiveness, and resistance to anoikis. VAL is undetectable in some mesenchymal cells, suggesting cell-type specific effects, and is directly correlated with poor patient prognosis and cancer progression. RNA interference targeting VAL presents therapeutic potential, and VAL/Vimentin pathway components may serve as both biomarkers and drug targets in aggressive cancer[1][2][4][5].
