Overview

Long intergenic non-protein coding RNA 1569 (LINC01569) is a human long non-coding RNA (lncRNA), located at 16p13.3, without protein-coding potential[1][3]. LINC01569 regulates gene expression and cellular functions by acting as a molecular sponge for microRNAs, thereby influencing downstream gene expression. In cancer biology, LINC01569 plays divergent roles depending on context: - In triple-negative breast cancer (TNBC), LINC01569 is downregulated and acts as a tumor suppressor, inhibiting tumor cell proliferation and migration by sponging miR-300, which in turn upregulates filamin A-interacting protein 1-like (FILIP1L)[2]. Reduced LINC01569 expression in TNBC is associated with more advanced tumors and poorer prognosis. - In colorectal cancer (CRC), LINC01569 is upregulated and promotes tumor proliferation, migration, and invasion by interacting with miR-381-3p and modulating RAP2A expression[5]. LINC01569 does not currently have known small-molecule or biologic drugs targeting it directly, but its expression level and regulatory interactions suggest roles as a diagnostic or prognostic biomarker in certain cancers. Its mechanism centers upon post-transcriptional regulation as a ceRNA, altering cancer cell behavior through the miRNA axis. No direct safety or therapeutic concerns have been reported, though manipulation of lncRNAs as therapeutic strategies remains an emerging field[2][5].

Mechanism of action

Not established for drugs; as a biomolecule, acts via sponging specific microRNAs (miRNA-300 in TNBC, miRNA-381-3p in CRC) which subsequently regulate downstream effectors such as filamin A-interacting protein 1-like (FILIP1L) and RAP2A
