Overview

LINC01572 is a cytoplasm-enriched, long non-coding RNA molecule (>200 nucleotides) that regulates gene expression post-transcriptionally. In cancer, it acts as an oncogenic factor by sponging tumor-suppressing microRNAs, upregulating glycolytic enzymes, and promoting pro-cancerous signaling pathways such as PI3K/AKT. Its overexpression correlates with advanced disease stage, high glucose-related markers (e.g., HbA1c in HCC with type 2 diabetes), and poor survival across various cancers. Knockdown of LINC01572 results in reduced proliferation, migration, clone formation, and increased ferroptosis, implying its role in cell survival and tumorigenesis. Its clinical importance lies in its emerging role as both a biomarker for aggressive disease and a potential therapeutic target in oncology

Mechanism of action

Acts as a competitive endogenous RNA (ceRNA), sponging miR-195-5p and upregulating PFKFB4 expression, thereby enhancing glycolysis and activating PI3K/AKT signaling. Promotes cancer cell survival and proliferation via deregulating cell cycle genes and suppressing ferroptosis (cell death due to iron-dependent lipid peroxidation). Facilitates β-catenin mRNA nuclear exportation in TNBC, promoting progression.
