Overview

Long intergenic non-protein coding RNA 1606 (LINC01606) is a long noncoding RNA (lncRNA) longer than 200 nucleotides that does not encode a protein, but rather exerts regulatory effects at the RNA level[2][3]. LINC01606 is frequently upregulated in colon cancer and multiple myeloma, where it functions as an oncogene[2][1]. It promotes cancer cell stemness, proliferation, migration, and invasion. Mechanistically, in colon cancer, LINC01606 inhibits ferroptosis (a form of regulated cell death driven by iron and lipid peroxidation) by modulating lipid metabolism through the SCD1–Wnt/β-catenin–TFE3 pathway. LINC01606 acts as a competing endogenous RNA for miR-423-5p, leading to the activation of canonical Wnt/β-catenin signaling and stemness-related genes such as CD133, CD44, Nanog, Epcam, Sox2, and Lgr5[2]. Its high expression is associated with poor prognosis and serves as a potential diagnostic and therapeutic biomarker, particularly in colon cancer and multiple myeloma[2][1]. No interacting drugs or direct therapeutic interventions have yet been clinically validated against LINC01606, but it is considered a promising cancer therapeutic target[2][1].
