Overview

Long intergenic non-protein coding RNA 1612 (LINC01612) is a cytoplasm-localized long noncoding RNA that functions primarily as a tumor suppressor in hepatocellular carcinoma (HCC)[1][2][4]. LINC01612 is significantly downregulated in HCC tissues compared to non-tumorous tissues and lower expression is correlated with poor pathological grading and worse prognosis[1][2][4]. Mechanistically, in p53-expressing hepatoma cells, LINC01612 acts as a competitive endogenous RNA by sponging miR-494 to increase ATF3 expression, which inhibits MDM2-mediated ubiquitination of p53, thus activating the p53 tumor suppressor pathway[1][2][4]. In p53-null hepatoma cells, LINC01612 interacts with and promotes the ubiquitin-mediated degradation of the oncoprotein YBX1, suppressing oncogenic behaviors[1][2][4]. These pathways collectively inhibit the growth and metastasis of hepatoma cells both in vitro and in vivo, supporting the role of LINC01612 as a functional lncRNA in liver cancer and a potential diagnostic biomarker and therapeutic target for HCC[1][2][4]. No drugs are currently known to directly target LINC01612, and no safety concerns or specific mechanisms of drug action are reported for this molecule.
