Overview

Long intergenic non-protein coding RNA 1638 (LINC01638) is a human long non-coding RNA gene product found on chromosome 22. It is highly expressed in human mesenchymal stem cells, where it maintains cell proliferation, cell cycle progression, and pluripotency. Experimental knockdown of LINC01638 in stem cells impairs osteogenic differentiation, induces DNA damage, cell senescence, and cell death. In cancer biology, LINC01638 is overexpressed in HER2-positive breast cancer, hepatocellular carcinoma, bladder cancer, and triple-negative breast cancer. It promotes tumor proliferation, prevents apoptosis, and drives metastasis by epigenetic regulation of cancer-related genes such as DNMTs, GLUT1, and ROCK2. High LINC01638 expression correlates with poor prognosis and increased risk of recurrence in cancers such as HER2-positive breast cancer and postoperative bladder cancer. As of current literature, LINC01638 has not been directly targeted by approved drugs, but is considered a promising candidate for cancer diagnosis and therapeutic intervention.
