Overview

LINC00665 is a long intergenic non-coding RNA gene located on chromosome 19q13.12, consisting of 1,749 bp and eight exons. It is primarily expressed in the cytosol but is also found in the nucleus and cytoskeleton. LINC00665 acts as an oncogenic non-coding RNA in many human cancers, where it is typically overexpressed relative to normal tissue. It exerts its biological activities by regulating gene expression as a competing endogenous RNA (ceRNA), binding to microRNAs (such as miR-9-5p, miR-214-3p, miR-3619), and modulating crucial oncogenic signaling pathways (e.g., Wnt/β-catenin, PI3K/Akt, TGF-β, NF-κB). In some cases, it directly interacts with proteins or chromatin modifiers (e.g., EZH2, LSD1, PKR). Knockdown of LINC00665 impairs tumor cell proliferation, migration, invasion, and increases cell apoptosis and chemosensitivity. LINC00665 is under investigation as both a biomarker and a preclinical therapeutic target, but no approved drugs directly modulate its expression yet.

Mechanism of action

Knockdown/inhibition of LINC00665 sensitizes tumor cells to chemotherapy (cisplatin) by modulating endoplasmic reticulum stress and apoptosis. Modulation of microRNA sponging or interference with associated signaling pathways (Wnt/β-catenin, TGF-β, PI3K/Akt, etc.).
