Overview

Long intergenic non-protein coding RNA 1673 (LINC01673) is a long non-coding RNA gene located on chromosome 17 that lacks protein-coding potential. It plays important regulatory roles in gene expression by epigenetically silencing tumor suppressor genes and modulating key signaling pathways associated with cancer cell proliferation, migration, invasion, and epithelial-mesenchymal transition. LINC01673 acts as a molecular scaffold and as a competing endogenous RNA, regulating microRNA activity in a tissue- and context-dependent manner. While LINC01673 promotes tumor progression in most cancers, it may function as a tumor suppressor in pancreatic cancer. Its elevated expression is considered a poor prognostic indicator for several malignancies, and its presence in peripheral blood positions it as a candidate biomarker for diagnostic and prognostic applications. No drugs are currently known to specifically target LINC01673, although its mechanistic involvement with epigenetic enzymes and regulator proteins suggests therapeutic opportunities.

Mechanism of action

Regulation of tumor suppressor gene expression via recruitment of epigenetic repressors (e.g., EZH2, LSD1, DNMT1) leading to histone methylation and transcriptional silencing. Acting as a molecular sponge for microRNAs (miR-515-5p, miR-205, miR-150-5p, miR-504) to regulate target mRNA expression. Formation of scaffolds that influence signaling pathways (e.g., WNT/β-catenin, PI3K/AKT).

Biological functions

Regulation of gene expression through epigenetic mechanismsFormation of competitive endogenous RNA (ceRNA) networks with microRNAsRegulation of cell proliferationPromotion of epithelial-mesenchymal transition (EMT)Regulation of cell migration and invasionAnti-apoptosis functions
04

Disease associations

Cancer (including non-small-cell lung cancer, gastric cancer, glioma, thyroid carcinoma, breast cancer, pancreatic cancer, melanoma)
05

Safety considerations

No direct safety or toxicity concerns identified, but the technical challenge of accurately detecting circulating levels in blood remains
06

Biomarkers

High expression in tumor tissue and peripheral blood linked to advanced disease and poor prognosis in several cancersPotential biomarker for prognosis, monitoring tumor development, and evaluating therapeutic efficacy
