Long intergenic non-protein coding RNA 1714 (LINC01714) is a bona fide long non-coding RNA that does not encode a protein and has been identified as a tumor suppressor specifically downregulated in cholangiocarcinoma (CCA) and some liver cancers[1]. LINC01714 acts partly by physically interacting with the FOXO3 protein, stabilizing it and inhibiting its phosphorylation at Ser318, which in turn suppresses cell proliferation, migration, and invasion. LINC01714 knockdown is associated with upregulation of pathways involved in tumor growth and metastasis, including the insulin signaling, VEGF, and MAPK pathways. Clinically, reduced LINC01714 expression or copy number predicts poorer patient survival in CCA. Importantly, increased LINC01714 expression sensitizes CCA cells to gemcitabine treatment, highlighting its potential as both a prognostic biomarker and a therapeutic target in combination chemotherapy for cholangiocarcinoma[1].
LINC01714 enhances sensitivity to gemcitabine by inhibiting FOXO3-Ser318 phosphorylation, thereby modulating FOXO3 protein activity[1].
