Overview

Long intergenic non-protein coding RNA 1811 (LINC01811) is a lncRNA highly expressed in colorectal cancer tissues and cell lines compared to normal tissue. It functions as an oncogenic factor primarily by sponging miR-214-3p, thereby increasing *YAP1* expression and promoting cell migration, invasion, and epithelial-mesenchymal transition. LINC01811 silencing reduces metastatic potential and alters mRNA/protein levels of EMT-related markers (such as vimentin, MMP2, MMP9, and E-cadherin). Its role as a ceRNA highlights its potential as a biomarker and a novel therapeutic target in colorectal cancer. There are currently no drugs directly targeting LINC01811, but its regulatory axis (LINC01811/miR-214-3p/YAP1) presents a promising area for future drug development.

Mechanism of action

LINC01811 acts by sponging microRNA (miR-214-3p), which leads to increased expression of *YAP1*, a known oncogenic factor in CRC progression. Targeting LINC01811 may inhibit invasion and migration of cancer cells by reversing EMT and reducing matrix metalloproteinase activity (MMP2, MMP9).
