Overview

Long intergenic non-protein coding RNA 1812 (LINC01812) is a long non-coding RNA localized on chromosome 2p14, functioning primarily as a regulator within the tumor microenvironment[3]. LINC01812 is overexpressed in cholangiocarcinoma and packaged into tumor-derived exosomes. It is internalized by macrophages, where it induces M2 polarization via the PI3K/AKT/mTOR pathway, promoting tumor cell perineural invasion and metastasis. This activity is achieved through its action as a ceRNA, sponging miR-101-3p, which relieves the repression on EZH2, an oncogenic epigenetic modifier. Elevated exosomal LINC01812 correlates with poor clinical prognosis and increased tumor invasiveness, establishing it as a promising biomarker and a potential therapeutic target in cancer, particularly cholangiocarcinoma[2][4][6]. LINC01812 exemplifies the regulatory and disease-modifying capacity of long intergenic non-coding RNAs in cancer biology[7].

Mechanism of action

Functions as a competing endogenous RNA (ceRNA), sponging miR-101-3p, thereby leading to increased EZH2 expression and activation of PI3K/AKT/mTOR pathway, which results in M2 macrophage polarization and perineural invasion[2][4]. Activates Notch signaling in macrophages[2].
