Overview

Long intergenic non-protein coding RNA 184 (LINC00184) is a human long non-coding RNA (lncRNA) gene, located at chromosome 1q42, and classified as an intergenic lncRNA since it does not overlap with protein-coding loci[5][9]. LINC00184 is highly expressed in several cancer types, including esophageal cancer, where its upregulation is associated with enhanced tumor cell proliferation, migration, invasion, and glycolytic metabolism[3][7]. Mechanistically, LINC00184 acts as an epigenetic regulator by recruiting DNA methyltransferase 1 (DNMT1) to methylate the promoter of the tumor suppressor PTEN, resulting in decreased PTEN expression, increased Akt pathway activity, and promotion of glycolysis in cancer cells[3]. Silencing of LINC00184 reverses these effects, suppressing tumor growth and restoring mitochondrial respiratory function, suggesting it is a potential therapeutic target and biomarker for cancer progression and prognosis[3]. No small-molecule drugs or direct inhibitors of LINC00184 are currently reported, and no clinical safety data are available. LINC00184 is an attractive therapeutic and biomarker candidate due to its regulatory role in cancer cell metabolism and epigenetics, but research is preclinical to date[3][7][9].
