Overview

LINC01852 (Long intergenic non-protein coding RNA 1852) is a long non-coding RNA found to be downregulated in colorectal cancer. Functional studies demonstrate that LINC01852 suppresses tumorigenesis and chemoresistance by binding serine/arginine-rich splicing factor 5 (SRSF5) and promoting its degradation via TRIM72-mediated ubiquitination, which inhibits SRSF5-dependent alternative splicing of the pyruvate kinase M (PKM) gene, shifting metabolism from glycolysis to oxidative phosphorylation. These effects attenuate cancer cell growth and resistance to chemotherapy, positioning LINC01852 as a promising therapeutic and prognostic biomarker in CRC[2][3][5].

Mechanism of action

When targeted (e.g., overexpressed experimentally), LINC01852 inhibits tumorigenesis and chemoresistance by suppressing SRSF5-mediated alternative splicing of PKM, therefore reducing PKM2-dependent glycolysis and promoting oxidative phosphorylation in CRC cells

03

Biological functions

Regulation of alternative splicing (specifically suppressing SRSF5-mediated alternative splicing of PKM)Inhibition of aerobic glycolysis (by decreasing PKM2 production)Modulation of ubiquitination and protein degradation (promotes TRIM72-mediated ubiquitination of SRSF5)Negative regulation of cell proliferationChemoresistance attenuation
04

Disease associations

Cancer (specifically colorectal cancer)Chemoresistance (in CRC)
05

Safety considerations

No clinical studies reported; unknown safety concerns for therapeutic manipulation in humans
