Overview

Long intergenic non-protein coding RNA 1918 (LINC01918), known as ROSALIND, is a recently discovered nuclear-encoded lincRNA that localizes to the mitochondrial matrix, where it interacts with mitochondrial ribosomes[2][4][6]. It features a high guanine (G) content, making it unusually susceptible to oxidative modifications. ROSALIND acts as a substrate for mitochondrial ROS, effectively buffering oxidative stress and thus protecting mitochondrial translation machinery from ROS-induced damage. Inhibiting ROSALIND expression leads to increased ROS and protein oxidation, impaired mitochondrial respiration, reduced cancer cell viability, and heightened immunogenicity. Upregulation of ROSALIND is seen in drug-tolerant and recurrent tumors, and its expression can stratify patients by response to immune checkpoint blockade in melanoma. This highlights ROSALIND’s role as a mitochondrial ROS-buffering system and as an emerging marker and potential target in cancer biology[2][4][6].
