Overview

Long intergenic non-protein coding RNA 210 (LINC00210) is a long non-coding RNA (lncRNA), more specifically a long intergenic non-coding RNA (lincRNA), with no known protein-coding potential[1][5]. It is frequently copy number gained and highly expressed in liver tumor-initiating cells (TICs) and hepatocellular carcinoma[2][4]. LINC00210 promotes the self-renewal capacity and tumorigenicity of liver TICs by activating the Wnt/β-catenin signaling pathway. Mechanistically, LINC00210 interacts with CTNNBIP1 (β-catenin interacting protein 1), inhibiting its suppression of β-catenin, thus enabling β-catenin to stimulate transcription of Wnt target genes. This pathway is implicated in stemness, metastasis, and treatment resistance in liver cancer. Targeting the LINC00210–CTNNBIP1–Wnt/β-catenin axis is under investigation as a potential therapeutic approach for eliminating liver TICs and combating hepatocellular carcinoma progression[2][4]. There is no evidence it is a receptor, enzyme, or transporter; its primary mode of action is as an RNA regulatory factor.

Mechanism of action

In experimental systems, knockdown (e.g., via antisense oligonucleotides or CRISPRi) suppresses LINC00210 activity, leading to attenuation of Wnt/β-catenin signaling and reduced tumorigenic potential; inhibition of LINC00210 may disrupt the LINC00210–CTNNBIP1 interaction and restore inhibitory regulation of β-catenin
