Long intergenic non-protein coding RNA 222 (LINC00222) is a non-coding RNA transcript longer than 200 nucleotides that does not encode proteins, classified as a lincRNA because it is transcribed from intergenic regions. LINC00222 has been characterized in lung adenocarcinoma, where its expression is frequently downregulated. Experimental overexpression of LINC00222 inhibits cancer cell proliferation, migration, and invasion, and induces cell apoptosis, partly by promoting the activity of glycogen synthase kinase-3β (GSK3β), a key negative regulator of the Wnt signaling pathway. These properties highlight its potential role as a tumor suppressor and promising biomarker. While LINC00222 may play regulatory roles in other cancers or disease processes, it is not currently targeted by known drugs, and functional characterization is ongoing.
Drugs or molecular therapies that increase LINC00222 expression might inhibit proliferation and metastasis, and promote apoptosis of cancer cells via activation of GSK3β and suppression of Wnt/β-catenin signaling
