Overview

Long intergenic non-protein coding RNA 2283 (LINC02283) is a cytoplasmic lncRNA gene that is co-amplified and highly expressed in glioblastoma stem-like cells and glioma patient samples with PDGFRA amplification or mutation. LINC02283 binds directly to the PDGFRA protein, enhancing its phosphorylation and activating its downstream AKT and ERK signaling cascades, thus promoting cell proliferation and tumorigenesis. Knockdown or inhibition of LINC02283 by antisense oligonucleotides decreases PDGFRA-mediated oncogenic activities and attenuates tumor growth in xenograft models. Clinically, high LINC02283 expression is correlated with poor prognosis in PDGFRA-driven glioblastoma, and its activity makes it a potential target for future RNA-based therapies. LINC02283 is not a classic receptor, enzyme, ion channel, or transporter; it is a noncoding RNA acting as a scaffold/regulator for protein activity—part of the rapidly emerging therapeutic RNA landscape.

Mechanism of action

Antisense oligonucleotide mediated knockdown—leads to attenuation of LINC02283 function, reduced PDGFRA signaling, decreased cell proliferation, and extended survival in glioblastoma models. Potential indirect modulation by PDGFRA inhibitors (e.g., AG1296), which may abrogate the enhanced signaling induced by LINC02283 overexpression.
