Long intergenic non-protein coding RNA 2303 (LINC02303), also known as TP53-regulated modulator of p27 (TRMP), is a human long noncoding RNA implicated in the regulation of cell proliferation and the cell cycle, especially in cancer cells[1][2][4][5]. It acts as a downstream effector in the TP53 (p53) pathway: induced by p53, TRMP and its shorter isoform TRMP-S function to suppress the expression of the cyclin-dependent kinase inhibitor p27 through multiple mechanisms. These include direct interaction with the RNA-binding protein PTBP1, competition for p27 mRNA binding, regulation of UHRF1 stability (an epigenetic repressor of p27), and translational repression of p53 via sequestration of RPL26, ultimately tuning p27 protein levels[2][4]. By repressing p27, TRMP/TRMP-S promote cell proliferation and inhibit cellular senescence, and their dysregulation has been linked to tumorigenic processes, highlighting their potential as candidate biomarkers or future targets of cancer therapy research, though they are not established as direct drug targets at this time[2][4][5].
