Long intergenic non-protein coding RNA 2320 (LINC02320) is a long non-coding RNA located on chromosome 14 and primarily classified as a lincRNA. It is highly expressed in colorectal cancer, especially metastatic lesions. LINC02320 promotes cancer cell proliferation, migration, and metastasis while inhibiting apoptosis. Mechanistically, it acts as a molecular scaffold in the nucleus, recruiting the transcription regulator ILF2 to the GRB7 gene promoter, thus activating GRB7 transcription and the MAPK signaling pathway. This pathway enhances downstream signaling, facilitating tumor growth and forming a positive feedback loop with the transcription factor FOS, which further upregulates LINC02320. Therapeutically, antisense oligonucleotides targeting LINC02320 have shown efficacy in blocking tumor growth in preclinical models, identifying it as a promising target for intervention in metastatic colorectal cancer
