Overview

Long intergenic non-protein coding RNA 2381 (LINC02381, also called lncEPAT, EGFR pathway-activated transcript, HOXC10 mRNA stabilizing factor, and LOC400043) is a human long non-coding RNA gene located on chromosome 12q13, downstream of the HOXC gene cluster[2][7]. LINC02381 is upregulated in various malignancies, including glioma and breast cancer, and also participates in endometriosis pathogenesis[1][5]. Mechanistically, LINC02381 acts as a competitive endogenous RNA, sponging microRNAs such as miR-27b-3p (in endometriosis) and miR-1271-5p (in breast cancer), thereby upregulating target genes (e.g., CTNNB1 or FN1) that promote cell proliferation, invasion, and migration[1][5]. Its expression is regulated by the EGFR pathway, with increased levels seen in tumors exhibiting EGFR amplification or activation[2]. LINC02381's functions have positioned it as a candidate biomarker and putative therapeutic target in diseases characterized by dysregulated cell growth and invasion[1][2][5].

Mechanism of action

No direct drug-targeting mechanisms described in the literature. Modulation of LINC02381 could theoretically affect tumorigenesis and proliferation via: - miRNA sponging (e.g., miR-27b-3p, miR-1271-5p) - Regulation of EGFR-mediated signaling pathways

Safety considerations

There are no direct therapeutic interventions yet; thus, specific safety concerns are not described for targeting LINC02381. Caution would be warranted for potential off-target effects typical for RNA-based therapies (e.g. siRNA, antisense oligonucleotides). As an lncRNA, its tissue- and disease-specific expression might minimize systemic risks, but this requires experimental validation.

Biomarkers

Expression of LINC02381 itself as a biomarker for disease state (glioma grade, breast cancer progression, endometriosis)
