Long intergenic non-protein coding RNA 2428 (LINC02428) is a long noncoding RNA highly expressed in normal liver tissue but downregulated in hepatocellular carcinoma (HCC)[1][2]. LINC02428 functions as a tumor suppressor by inhibiting proliferation, migration, and metastasis of HCC cells both in vitro and in vivo[1][2]. Mechanistically, LINC02428 binds to insulin-like growth factor-2 mRNA-binding protein 1 (IGF2BP1) in the cytoplasm, which prevents IGF2BP1 from stabilizing lysine demethylase 5B (KDM5B) mRNA, thereby decreasing KDM5B expression and disrupting a pro-tumorigenic positive feedback loop between KDM5B and IGF2BP1[1][2]. Low LINC02428 expression in HCC is associated with poor prognosis, and its expression may serve as a biomarker for prognosis in liver cancer patients[1][2]. No interacting drugs or direct mechanisms of drug action have been described, and there are currently no documented therapeutic safety concerns related specifically to targeting LINC02428.
