Overview

Long intergenic non-protein coding RNA 2568 (LINC02568) is a long non-coding RNA that is highly expressed in estrogen receptor (ER)-positive breast cancer. It functions both in the cytoplasm—where it acts as a competing endogenous RNA (ceRNA) to stabilize ESR1 (estrogen receptor alpha) mRNA through miRNA sponging (notably of miR-1233-5p and miR-874-3p)—and in the nucleus, where it regulates the transcription of adjacent gene CA12, thereby contributing to tumor-specific pH homeostasis. LINC02568 promotes tumorigenic activities such as cell proliferation, migration, invasion, and confers resistance to endocrine therapy. Its inhibition reduces tumor growth and synergistically enhances the effects of both endocrine therapy and CA12 inhibition. This makes LINC02568 an emerging therapeutic target and potential biomarker for prognosis and drug resistance in ER-positive breast cancer[2][3][4].

Mechanism of action

Antisense oligonucleotides inhibit LINC02568 to decrease ER-positive breast cancer growth and tumorigenesis by destabilizing ESR1 mRNA and reducing estrogen receptor signaling. Combined inhibition (LINC02568 + endocrine therapy, or LINC02568 + CA12 inhibitor) has a synergistic antitumor effect.
