Overview

Long intergenic non-protein coding RNA 2605 (LINC02605) is a nuclear long non-coding RNA that is induced by viral infection and type I interferon stimulation in an NF-κB and JAK-STAT dependent manner[1][3]. LINC02605 acts as a positive regulator of the antiviral innate immune response by promoting the nuclear translocation of interferon regulatory factor 3 (IRF3), leading to increased type I interferon (IFN-α/β) production and inhibition of viral replication[1][3]. Mechanistically, it functions by binding to hsa-miR-107, thus relieving the inhibition on phosphatase and tensin homolog (PTEN) expression, which subsequently promotes dephosphorylation and nuclear import of IRF3[1][3]. Knockdown of LINC02605 leads to reduced PTEN expression, decreased antiviral gene activation, and enhanced viral replication, while overexpression has the opposite effect[1][3]. Beyond antiviral immunity, LINC02605/alias RP11-79H23.3 is downregulated in bladder cancer and may inhibit tumor progression by upregulating PTEN and suppressing PI3K/Akt signaling[1]. LINC02605 does not code for a protein and has no reported direct drug interactions or known biomarker or safety issue associations to date[1][3].
