Long intergenic non-protein coding RNA 2774 (LINC02774) is a nuclear long non-coding RNA primarily studied in the context of glioma, a type of brain tumor. It functions as a tumor suppressor: overexpression of LINC02774 reduces glioma cell proliferation, colony formation, invasion, migration, and in vivo tumor growth, while its knockdown has the opposite effects. Mechanistically, LINC02774 interacts with its neighboring gene RP58 (ZBTB18), resulting in enhanced expression of the enzyme PHD3, which catalyzes hydroxylation and thereby promotes the degradation of the transcription factor HIF-1α. This pathway results in decreased levels of glycolysis-promoting genes, such as VEGF and GLUT1, effectively inhibiting glycolysis. LINC02774 expression negatively correlates with glioma malignancy and may serve as a valuable prognostic biomarker and potential therapeutic target for glioma, but no interacting drugs or associated safety concerns have yet been identified in the literature[1][2].
