Overview

Long intergenic non-protein coding RNA 2820 (LINC02820) is a long noncoding RNA distinctly upregulated in esophageal squamous cell carcinoma (ESCC) tissue versus normal tissue[1][2][5]. It is present in both the nucleus and cytoplasm of ESCC cells and typically consists of three exons for a total length of about 653–695 nucleotides[1]. LINC02820 mainly acts as an oncogene, promoting cell proliferation, migration, invasion, tumor growth, and metastasis both in vitro and in vivo[1][2][4][5]. Mechanistically, it achieves this by binding to and stabilizing the MYH9 protein, protecting it from ubiquitin-mediated proteasomal degradation, as well as being stabilized itself by the RNA-binding protein IGF2BP2[1][4]. It can also interact with splice factor 3B subunit 3 (SF3B3) and, importantly, cooperates with TNFα to amplify the NF-κB signaling pathway, enhancing cytoskeletal remodeling and driving metastatic processes in ESCC[2][5]. These findings indicate LINC02820 is a potential therapeutic target and biomarker in ESCC, although no clinically approved drugs currently target it, and its full safety profile is unclear.
