Long intergenic non-protein coding RNA 324 (LINC00324) is a cytoplasm-localized long noncoding RNA, approximately 2 kb in length, generally upregulated in a variety of cancers including gastric cancer, lung cancer, hepatocellular carcinoma, nasopharyngeal carcinoma, and others. LINC00324 exerts effects predominantly through three molecular mechanisms: acting as a competing endogenous RNA (ceRNA) to sponge tumor-suppressive miRNAs; binding with RNA-binding proteins such as HuR to stabilize oncogenic mRNAs (e.g., FAM83B, WDR66); and recruiting transcription factors like PU.1 to activate the transcription of target genes (e.g., FasL in liver cancer). LINC00324 is frequently associated with enhanced cell proliferation, migration, invasion, metastasis, and resistance to apoptosis in cancer. It is considered a prognostic biomarker and an emerging RNA-based therapeutic target, though as of now, there are no approved drugs directly targeting LINC00324.
